Anti-Cancer N-Heterocyclic Carbene Complexes of Gold: Thiol “Switch-on” Fluorescent Probes, Thioredoxin Reductase Inhibitors and Endoplasmic Reticulum Targeting Agents (Springer Theses)

This thesis focuses on the development of gold- and non-classical platinum-based anti-cancer agents that display distinctively different anti-cancer mechanisms compared to the commonly used cisplatin. These metal complexes contain N-heterocyclic carbene (NHC) ligands which are able to form strong M-C(NHC) bonds, conferring high stability and favorable lipophilicity, reactivity and binding specificity of metal complexes on biomolecules. The author demonstrates significant advances made in anti-cancer gold(III), gold(I) and platinum(II) complexes. Detailed chemical synthesis, in vitro and/or in vivo anti-cancer activities are clearly presented including: (i) a class of Au(III) complexes containing a highly fluorescent N^N^N ligand and NHC ligand that simultaneously act as fluorescent thiol "switch-on" probes and anti-cancer agents; (ii) a dinuclear gold(I) complex with a mixed diphosphine and bis(NHC) ligand displaying favorable stability and showing significant inhibition of tumor growth in two independent mice models with no observable side effects; and (iii) a panel of stable luminescent cyclometalated platinum(II) complexes exhibiting high specificity to localize to the endoplasmic reticulum (ER) domain, inducing ER stress and cell apoptosis. These works highlight the clinical potential that gold and platinum complexes offer for cancer treatment.

The Language of Trauma: War and Technology in Hoffmann, Freud, and Kafka

From the Napoleonic Wars to the invention of the railway to the shell shock of World War I, writers tried to give voice to the suffering that war and industrial technology had wrought all around them. Yet they, like the doctors who treated these victims, repeatedly ran up against the incapacity of language to describe such anguish; those who suffered trauma, those who tried to heal it, and those who represented it were all unable to find the appropriate words. In The Language of Trauma, John Zilcosky uncovers the reactions of three major central European writers – E.T.A. Hoffmann, Sigmund Freud, and Franz Kafka – to the birth of modern trauma in the nineteenth and early twentieth centuries. Zilcosky makes the case that Hoffmann, Freud, and Kafka managed to find the language of trauma precisely by not attempting to name the trauma conclusively and instead allowing their writing to mimic the experience itself. Just as the victims’ symptoms seemed not to correspond to a physical cause, the writers’ words did not connect directly to the objects of the world. While doctors attempted to overcome this indeterminacy, these writers embraced and investigated it; they sought a language that described language’s tragic limits and that, in so doing, exemplified the wider literary and philosophical crisis of their time. Zilcosky boldly argues that this linguistic scepticism emerged together with the medical inability to name the experience of trauma. He thereby places trauma where it belongs: at the heart of both medicine’s diagnostic predicament and modern literature’s most daring experiments.

Growth factor receptors have long been known to drive malignant transformation and cancer progression. The epidermal growth factor receptor (EGFR, ErbB, HER) system is likely the best described membrane receptor tyrosine kinase family in malignant tumors. With implementation of the growth-inhibitory anti-HER-2 antibody trastuzumab (Herceptin) for the treatment of HER-2-positive advanced metastatic breast cancer, a new era has dawned in the therapy of this malignant disease. Unfortunately, trastuzumab-sensitive cancers invariably develop resistance to the antibody after some time. Recent clinical studies have revealed that these refractory tumors are still responsive to inhibition of the HER receptor family using dual HER-1/-2 inhibitors such as lapatinib (Tykerb/Tyverb). Moreover, a multiplicity of novel, improved irreversibly acting small molecular HER tyrosine kinase inhibitors are in the pipeline of many drug developing companies and are being evaluated in the clinical setting.
